Head over to Tinker's Nook in Cottonpuff Field and ask her if she has any quests. If you have finished the quest, Restocking Tinker's Nook, the quest should appear in the list of quests you may do and you may begin immediately. Upon starting the quest, Tink will greet you:

Tinker Bell: "Oh, Pixie Name Here, I feel awful. It's my tinker's hammer -- it's worn out again! I suppose I shouldn't be upset, though. I mean, I use it all the time. No wonder it wears out!"
"I could use your help fixing it. What do you say?"

Step 1

Accept this Quest?

Once you agree, Tink is thrilled and gives you the first step.

Tinker Bell: "Thanks! Now, for this job I need special glue. Not just any glue is strong enough to repair a tinker's hammer, you know."
"The first ingredient is Spider Silk. Will you go get some?"

Gather 15 pieces of Spider Silk and bring them to Tinker's Nook in Cottonpuff Field.
